Characterization of Chromatin Accessibility in Fetal Bovine Chondrocytes

Despite significant advances of the bovine epigenome investigation, new evidence for epigenetic basis fetal cartilage development remains lacking. In this study, chondrocytes were isolated from long bone tissues fetuses at 90 days. The Assay Transposase-Accessible Chromatin with high throughput sequencing (ATAC-seq) and transcriptome (RNA-seq) used to characterize gene expression chromatin accessibility profile in chondrocytes. A total 9686 open regions identified 45% peaks enriched promoter regions. Then, all annotated nearest Gene Ontology (GO) Kyoto Encylopaedia Genes Genomes (KEGG) analysis. Growth development-related processes such as amide biosynthesis process (GO: 0043604) translation regulation 006417) GO KEGG analysis endoplasmic reticulum protein processing signal pathway, TGF-β signaling pathway cell cycle which are closely related synthesis during proliferation. Active transcription factors (TFs) by ATAC-seq, fully verified levels obtained RNA-seq. Among top50 TFs footprint analysis, known or potential FOS, FOSL2 NFY found. Overall, our data provide a theoretical further determining regulatory mechanism bovine.
